It is not desirable to eject the phone is turned ON this will cause some applications to force close and such that if they are accessing the Memory card. There is one way you can remove the card when the phone is turned ON all you have to do is open the "Settings" menu from the application drawer and then select the option as "Storage" select that there you will find an option as "Unmount SD card" select that so that the SD card will be Unmounted and so you will be able to remove it without having any errors.

Yes it should be done that way. The eject option is given to safely remove your device without hampering the data whatever is inside the sd card.
After you safely eject it you can remove your sd card. Again whenever you want to insert it again you can do that. None of the information will get lost it will be the same as before.
